RESUMEN

This study conducted a spatio-temporal analysis of runoff, total suspended sediment, suspended particulate carbon, nitrogen, and phosphorus loadings within the 2.06 km2 Steppler subwatershed in southern Manitoba of Canada based on 11 years of field monitoring data collected at nine stations. Results showed that the nutrient losses were very small because of the implementation of multiple BMPs in the study area. However, a high spatio-temporal variation of runoff and water quality parameters was found for the nine fields within the subwatershed. The average runoff coefficient was 0.19 at the subwatershed outlet with sediment, suspended particulate carbon, total nitrogen, and total phosphorus losses of 73.8, 6.10, 4.54, and 0.76 kg/ha respectively. Spring snowmelt runoff was about 74.5% of the annual runoff at the subwatershed outlet, while for sediment, suspended particulate carbon, total nitrogen, and total phosphorus, the proportions were 61.1%, 63.6%, 74.9%, and 81.2% respectively during the monitoring period, which suggests that BMPs designed for reducing nutrient loadings from snowmelt runoff would be more effective than BMPs designed for reducing pollutant loading from rainfall storms in the study area. Research findings from this study will benefit the enhancement of current BMPs and the development of new BMPs in the region to minimize soil and nutrient losses from agricultural fields and improve water quality in receiving water bodies.

On 11th March 2020, the UK government announced plans for the scaling of COVID-19 testing, and on 27th March 2020 it was announced that a new alliance of private sector and academic collaborative laboratories were being created to generate the testing capacity required. The Cambridge COVID-19 Testing Centre (CCTC) was established during April 2020 through collaboration between AstraZeneca, GlaxoSmithKline, and the University of Cambridge, with Charles River Laboratories joining the collaboration at the end of July 2020. The CCTC lab operation focussed on the optimised use of automation, introduction of novel technologies and process modelling to enable a testing capacity of 22,000 tests per day. Here we describe the optimisation of the laboratory process through the continued exploitation of internal performance metrics, while introducing new technologies including the Heat Inactivation of clinical samples upon receipt into the laboratory and a Direct to PCR protocol that removed the requirement for the RNA extraction step. We anticipate that these methods will have value in driving continued efficiency and effectiveness within all large scale viral diagnostic testing laboratories.

We report the development of a large scale process for heat inactivation of clinical COVID-19 samples prior to laboratory processing for detection of SARS-CoV-2 by RT-qPCR. With more than 266 million confirmed cases, over 5.26 million deaths already recorded at the time of writing, COVID-19 continues to spread in many parts of the world. Consequently, mass testing for SARS-CoV-2 will remain at the forefront of the COVID-19 response and prevention for the near future. Due to biosafety considerations the standard testing process requires a significant amount of manual handling of patient samples within calibrated microbiological safety cabinets. This makes the process expensive, effects operator ergonomics and restricts testing to higher containment level laboratories. We have successfully modified the process by using industrial catering ovens for bulk heat inactivation of oropharyngeal/nasopharyngeal swab samples within their secondary containment packaging before processing in the lab to enable all subsequent activities to be performed in the open laboratory. As part of a validation process, we tested greater than 1200 clinical COVID-19 samples and showed less than 1 Cq loss in RT-qPCR test sensitivity. We also demonstrate the bulk heat inactivation protocol inactivates a murine surrogate of human SARS-CoV-2. Using bulk heat inactivation, the assay is no longer reliant on containment level 2 facilities and practices, which reduces cost, improves operator safety and ergonomics and makes the process scalable. In addition, heating as the sole method of virus inactivation is ideally suited to streamlined and more rapid workflows such as 'direct to PCR' assays that do not involve RNA extraction or chemical neutralisation methods.

Phosphorus (P) runoff from agricultural land plays a critical role in downstream water quality. This article summarizes P and sediment runoff data for both snowmelt and rainfall runoff from 30 arable fields in the Canadian provinces of Saskatchewan, Manitoba and Ontario. The data were collected from 216 site-years of field experiments, with climates ranging from semi-arid to humid and a wide range of field management practices. In the article, mean annual and seasonal (in terms of snowmelt and rain) precipitation inputs, runoff depths, and P and sediment concentrations and loads are presented, along with ranges of yearly values. In addition, information of field management and soil characteristics (e.g. soil type and soil Olsen P) is also presented for each field. The data have potential to be reused for national and international cross-region comparisons of P and sediment losses, constructing and validating decision-support models and tools for assessing and managing P losses in both snowmelt and rainfall runoff, and informing beneficial management practices to improve agricultural water quality. Interpretation of the data is found in "Phosphorus runoff from Canadian agricultural land: A cross-region synthesis of edge-of-field results" [1].

Immunotherapies and targeted therapies have revolutionised treatment of metastatic melanoma and improved survival rates. However, survivors treated with novel therapies are vulnerable to high levels of fear of cancer recurrence or progression (FCR). Existing FCR interventions have rarely been trialled in people with advanced cancer. The current study aimed to evaluate the acceptability and feasibility of Fear-Less: a stepped-care model to treat FCR in people with metastatic melanoma treated with immunotherapy or targeted therapy. Sixty-one outpatients with metastatic melanoma were screened using the Fear of Cancer Recurrence Inventory Short Form (FCRI-SF) and Fear of Progression Questionnaire Short Form (FoP-Q-SF). Survivors with subthreshold FCR were stratified to a self-management intervention while those with clinical levels of FCR were provided with an individual therapy, Conquer Fear. Survivor experience surveys and rescreening were administered post-intervention completion. Results indicated that Fear-Less was an acceptable and feasible FCR intervention. Results provided preliminary support for the potential impact of Fear-Less in reducing FCR. Fear-Less is a promising first step in providing an acceptable and feasible stepped-care model to treat FCR in survivors with metastatic disease.

Snowmelt runoff often comprises the majority of annual runoff in the Canadian Prairies and a significant proportion of total nutrient loss from agricultural land to surface water. Our objective was to determine the effect of agroecosystem management on snowmelt runoff and nutrient losses from a long-term field experiment at Swift Current, SK. Runoff quantity, nutrient concentrations, and loads were estimated after a change in management from conventionally tilled wheat ( L.)-fallow (Conv W-F) to no-till wheat-fallow and subsequently no-till wheat-pulse (NT W-F/LP) and to an organic system with a wheat-green manure rotation (Org W-GM). The conversion from conventional tillage practices to no-till increased snowmelt runoff likely due to snow trapping by standing stubble after summer fallow. Relatedly, runoff after no-till summer fallow had higher dissolved P losses (0.07 kg P ha). Replacing summer fallow with a pulse crop in the no-till rotation decreased snowmelt runoff losses and nutrient concentrations. The Org W-GM treatment had the lowest P loss after stubble (0.02 kg P ha) but had high dissolved P concentrations in snowmelt following the green manure (0.55 mg P L), suggesting a contribution from incorporated crop residues. In this semiarid climate with little runoff, dissolved reactive P and NO-N loads in snowmelt runoff were smaller than those reported elsewhere on the prairies (averaging <0.05 kg P ha yr, and <0.2 kg NO-N ha yr); however, the nutrient concentrations we observed, in particular for P, even without P fertilizer addition for organic production, question the practicality of agricultural management systems in this region meeting water quality guidelines.

Cold agricultural regions are important sites of global food production. This has contributed to widespread water quality degradation influenced by processes and hydrologic pathways that differ from warm region analogues. In cold regions, snowmelt is often a dominant period of nutrient loss. Freeze-thaw processes contribute to nutrient mobilization. Frozen ground can limit infiltration and interaction with soils, and minimal nutrient uptake during the nongrowing season may govern nutrient export from agricultural catchments. This paper reviews agronomic, biogeochemical, and hydrological characteristics of cold agricultural regions and synthesizes findings of 23 studies that are published in this special section, which provide new insights into nutrient cycling and hydrochemical processes, model developments, and the efficacy of different potentially beneficial management practices (BMPs) across varied cold regions. Growing evidence suggests the need to redefine optimum soil phosphorus levels and input regimes in cold regions to allow achievement of water quality targets while still supporting strong agricultural productivity. Practices should be considered through a regional and site-specific lens, due to potential interactions between climate, hydrology, vegetation, and soils, which influence the efficacy of nutrient, crop, water, and riparian buffer management. This leads to differing suitability of BMPs across varied cold agricultural regions. We propose a systematic approach (""), to achieve water quality objectives in variable and changing climates, which combines nutrient transport process onceptualization, nderstanding BMP functions, redicting effects of variability and change, onsideration of producer input and agronomic and environmental tradeoffs, practice daptation, nowledge mobilization, and valuation of water quality improvement.

Reducing eutrophication in surface water is a major environmental challenge in many countries around the world. In cold Canadian prairie agricultural regions, part of the eutrophication challenge arises during spring snowmelt when a significant portion of the total annual nutrient export occurs, and plant residues can act as a nutrient source instead of a sink. Although the total mass of nutrients released from various crop residues has been studied before, little research has been conducted to capture fine-timescale temporal dynamics of nutrient leaching from plant residues, and the processes have not been represented in water quality models. In this study, we measured the dynamics of P and N release from a cold-hardy perennial plant species, alfalfa ( L.), to meltwater after freeze-thaw through a controlled snowmelt experiment. Various winter conditions were simulated by exposing alfalfa residues to different numbers of freeze-thaw cycles (FTCs) of uniform magnitude prior to snowmelt. The monitored P and N dynamics showed that most nutrients were released during the initial stages of snowmelt (first 5 h) and that the magnitude of nutrient release was affected by the number of FTCs. A threshold of five FTCs was identified for a greater nutrient release, with plant residue contributing between 0.29 (NO) and 9 (PO) times more nutrients than snow. The monitored temporal dynamics of nutrient release were used to develop the first process-based predictive model controlled by three potentially measurable parameters that can be integrated into catchment water quality models to improve nutrient transport simulations during snowmelt.

Managing P export from agricultural land is critical to address freshwater eutrophication. However, soil P management, and options to draw down soil P have received little attention in snowmelt-dominated regions because of limited interaction between soil and snowmelt. Here, we assessed the impacts of soil P drawdown (reducing fertilizer P inputs combined with harvest removal) on soil Olsen P dynamics, runoff P concentrations, and crop yields from 1997 to 2014 in paired fields in Manitoba, Canada. We observed that Olsen P concentrations in the 0- to 5-cm soil layer were negatively correlated with the cumulative P depletion and declined rapidly at the onset of the drawdown practice (3.1 to 5.4 mg kg yr during 2007-2010). In both snowmelt runoff and rainfall runoff, concentrations of total dissolved P (TDP) were positively correlated with the concentrations of soil Olsen P. Soil P drawdown to low to moderate fertility levels significantly decreased mean annual flow-weighted TDP concentrations in snowmelt runoff from 0.60 to 0.30 mg L in the field with high initial soil P and from 1.17 to 0.42 mg L in the field with very high initial soil P. Declines in TDP concentration in rainfall runoff were greater. Critically, yields of wheat ( spp.) and canola ( L.) were not affected by soil P depletion. In conclusion, we demonstrate that relatively rapid reductions in P loads are achievable at the field scale via managing P inputs and soil P pools, highlighting a management opportunity that can maintain food security while improving water security in cold regions.

The use of cover crops and crop residues is a common strategy to mitigate sediment and nutrient losses from land to water. In cold climates, elevated dissolved P losses can occur associated with freeze-thaw of plant materials. Here, we review the impacts of cover crops and crop residues on dissolved P and total P loss in cold climates across ∼41 studies, exploring linkages between water-extractable P (WEP) in plant materials and P loss in surface runoff and subsurface drainage. Water-extractable P concentrations are influenced by plant type and freezing regimes. For example, WEP was greater in brassica cover crops than in non-brassicas, and increased with repeated freeze-thaw cycles. However, total P losses in surface runoff and subsurface drainage from cropped fields under cold climates were much lower than plant WEP, owing to retention of 45 to >99% of released P by soil. In cold climatic regions, cover crops and crop residues generally prevented soil erosion and loss of particle-bound P during nongrowing seasons in erodible landscapes but tended to elevate dissolved P loss in nonerodible soils. Their impact on total P loss was inconsistent across studies and complicated by soil, climate, and management factors. More research is needed to understand interactions between these factors and plant type that influence P loss, and to improve the assessment of crop contributions to P loss in field settings in cold climates. Further, tradeoffs between P loss and the control of sediment loss and N leaching by plants should be acknowledged.

In northern regions, a high proportion of annual runoff and phosphorus (P) export from cropland occurs with snowmelt. In this study, we analyze 57 site-years of field-scale snowmelt runoff data from 16 small watersheds draining fine-textured soils (clay or clay loam) in Manitoba, Canada. These fields were selected across gradients of soil P (2.4 to 26.7 mg kg, 0- to 15-cm Olsen P), tillage intensity (high frequency to long-term no-till), and fertilizer input. The strongest predictor of flow-weighted mean concentrations of total dissolved P (TDP) in snowmelt runoff was Olsen P in the top 5 cm of soil ( = 0.45, < 0.01). Residual variation in this relationship related positively to volumetric soil moisture and negatively to water yield. Although Olsen P levels were relatively consistent from year to year, suggesting control by long-term fertilization and tillage history, Olsen P stratification (ratio of 0-5/0-15 cm) increased with rates of fertilizer application. Particulate P (PP) comprised <34% of total P on average, and concentrations were not well predicted by soil or management characteristics. Loads of PP and TDP exported during snowmelt were primarily a function of water yield and size of accumulated snowpack; however, residual variation in the TDP relationship correlated positively with both soil moisture and Olsen P. Retention of runoff water on the landscape could reduce loads, but careful management of near-surface soil P is required to prevent snowmelt runoff losses of P at the source and to reduce the potential for the eutrophication of downstream aquatic ecosystems.

OBJECTIVES: Selection, Optimization, and Compensation (SOC) may contribute to successful aging by helping older people maximize well-being in the context of physical decline. To explore this hypothesis, and to investigate the potential for narrative analysis to improve understanding of SOC, we analyze interviews conducted with 15 members of the 6-Day Sample, a cohort of Scots born in 1936. METHOD: Interviewees were chosen based on their physical function and well-being scores. Interviews were analyzed to investigate "SOC talk," that is, older people's talk about SOC behaviors in everyday life. Types and amounts of SOC talk were quantified, and talk was narratively analyzed. We hypothesized that older people who engaged in more SOC talk would have higher well-being. RESULTS: Older people who engaged in high levels of SOC talk had high well-being despite low physical function. Those who engaged in little SOC talk had low well-being despite higher physical function. DISCUSSION: The concept of successful aging is valuable in part because of its narrative quality: One must strive to keep one's life story developing despite physical decline and other losses. We provide evidence, from the perspectives of older people themselves, of the ways in which SOC may play a role in that process.

BACKGROUND: Functional outcome measurement tools exist for individual diagnoses (eg, stroke), but no prospectively validated mobility measure is available for physical therapists' use across the breadth of acute hospital inpatients. The modified Iowa Level of Assistance Scale (mILOA), a scale measuring assistance required to achieve functional tasks, has demonstrated functional change in inpatients with orthopedic conditions and trauma, although its psychometric properties are unknown. OBJECTIVE: The aim of this study was to assess interrater reliability, known-groups validity, and responsiveness of the mILOA in acute hospital inpatients. DESIGN: This was a cohort, measurement-focused study. METHODS: Patients at a large teaching hospital in Melbourne, Australia, were recruited. One hundred fifty-two inpatients who were functionally stable across 5 clinical groups had an mILOA score calculated during 2 independent physical therapy sessions to assess interrater reliability. Known-groups validity ("ready for discharge"/"not ready for discharge") and responsiveness also were assessed. RESULTS: The mean age of participants in the reliability phase of the study was 62.5 years (SD=17.7). The interrater reliability was excellent (intraclass correlation coefficient [2,1]=.975; 95% confidence interval=.965, .982), with a mean difference between scores of -.270 and limits of agreement of ±5.64. The mILOA score displayed a mean difference between 2 known groups of 15.3 points. Responsiveness was demonstrated with a minimal detectable change of 5.8 points. LIMITATIONS: Participants were included in the study if able to give consent for themselves, thereby limiting generalizability. Construct validity was not assessed due to the lack of a gold standard. CONCLUSIONS: The mILOA has excellent interrater reliability and good known-groups validity and responsiveness to functional change across acute hospital inpatients with a variety of diagnoses. It may provide opportunities for physical therapists to collect a functional outcome measure to demonstrate the effectiveness of inpatient therapy and allow for benchmarking across institutions.

Soils are subject to varying degrees of direct or indirect human disturbance, constituting a major global change driver. Factoring out natural from direct and indirect human influence is not always straightforward, but some human activities have clear impacts. These include land-use change, land management and land degradation (erosion, compaction, sealing and salinization). The intensity of land use also exerts a great impact on soils, and soils are also subject to indirect impacts arising from human activity, such as acid deposition (sulphur and nitrogen) and heavy metal pollution. In this critical review, we report the state-of-the-art understanding of these global change pressures on soils, identify knowledge gaps and research challenges and highlight actions and policies to minimize adverse environmental impacts arising from these global change drivers. Soils are central to considerations of what constitutes sustainable intensification. Therefore, ensuring that vulnerable and high environmental value soils are considered when protecting important habitats and ecosystems, will help to reduce the pressure on land from global change drivers. To ensure that soils are protected as part of wider environmental efforts, a global soil resilience programme should be considered, to monitor, recover or sustain soil fertility and function, and to enhance the ecosystem services provided by soils. Soils cannot, and should not, be considered in isolation of the ecosystems that they underpin and vice versa. The role of soils in supporting ecosystems and natural capital needs greater recognition. The lasting legacy of the International Year of Soils in 2015 should be to put soils at the centre of policy supporting environmental protection and sustainable development.

Population-based, prospective longitudinal cohort studies are considering the issues surrounding returning findings to individuals as a result of genomic and other medical research studies. While guidance is being developed for clinical settings, the process is less clear for those conducting longitudinal research. This paper discusses work conducted on behalf of The UK Cohort and Longitudinal Study Enhancement Resource programme (CLOSER) to examine consent requirements, process considerations and specific examples of potential findings in the context of the 1958 British Birth cohort. Beyond deciding which findings to return, there are questions of whether re-consent is needed and the possible impact on the study, how the feedback process will be managed, and what resources are needed to support that process. Recommendations are made for actions a cohort study should consider taking when making vital decisions regarding returning findings. Any decisions need to be context-specific, arrived at transparently, communicated clearly, and in the best interests of both the participants and the study.

The APOE ε2/3/4 genotype has been associated with low-density lipoprotein cholesterol (LDL-C) and Alzheimer disease. However, evidence for associations with measures of cognitive performance in adults without dementia has been mixed, as it is for physical performance. Associations may also be evident in other genotypes implicated in LDL-C levels. As part of the Healthy Ageing across the Life Course (HALCyon) collaborative research programme, genotypic information was obtained for APOE ε2/3/4, rs515135 (APOB), rs2228671 (LDLR) and rs629301 (SORT1) from eight cohorts of adults aged between 44 and 90 + years. We investigated associations with four measures of cognitive (word recall, phonemic fluency, semantic fluency and search speed) and physical capability (grip strength, get up and go/walk speed, timed chair rises and ability to balance) using meta-analyses. Overall, little evidence for associations between any of the genotypes and measures of cognitive capability was observed (e.g. pooled beta for APOE ε4 effect on semantic fluency z score = -0.02; 95 % CI = -0.05 to 0.02; p value = 0.3; n = 18,796). However, there was borderline evidence within studies that negative effects of APOE ε4 on nonverbal ability measures become more apparent with age. Few genotypic associations were observed with physical capability measures. The findings from our large investigation of middle-aged to older adults in the general population suggest that effects of APOE on cognitive capability are at most modest and are domain- and age-specific, while APOE has little influence on physical capability. In addition, other LDL-C-related genotypes have little impact on these traits.

RESUMEN

There is now a body of evidence that demonstrates strong links between neighbourhood characteristics and mental health and wellbeing. There is an increasing interest in how this relationship varies for individuals of different ages. Understanding the link between neighbourhood and wellbeing for older adults is of particular significance, given the changing age structure of the population and the desire among policy makers and practitioners to promote healthy and active ageing. This paper provides further evidence on the nature and strength of the link between individual perceptions of neighbourhood belonging and mental wellbeing among those over age fifty using both qualitative and quantitative data from three British cohort studies. Between 2008 and 2011 quantitative data were collected from 10,312 cohort members, and 230 of them took part in qualitative biographical interviews. Quantitative analysis confirms that there is a moderate association between neighbourhood cohesion and wellbeing measured at the individual level in each of the three cohorts. This association persists after controlling for a range of covariates including personality. The association between neighbourhood cohesion and wellbeing is stronger for individuals in the older two cohorts than in the younger cohort. Using qualitative biographical interviews with 116 men and 114 women we illustrate how individuals talk about their sense of neighbourhood belonging. The importance of social participation as a mechanism for promoting neighbourhood belonging, and the use of age and life stage as characteristics to describe and define neighbours, is clear. In addition, the qualitative interviews point to the difficulties of using a short battery of questions to capture the varied and multi-dimensional nature of neighbourhood relations.

An 8-yr field-scale study, 2005 to 2012, investigated effects of agricultural land use on nutrient and sediment losses during snowmelt runoff from four treatment fields in southern Manitoba. In 2005, two fields with a long-term history of annual crop (AC) production were planted to perennial forage (PF), while two other fields were left in AC production. In 2009, the AC fields were converted to PF, while the PF fields were returned to AC. Runoff flow rates were monitored at the lower edge of the fields, and nutrient concentrations of runoff water were determined. The effects of AC and PF on selected variables were similar for the spatial (between-fields) and temporal (within-field) comparisons. The flow-weighted mean concentrations (FWMCs) and loads of particulate N, P, and sediment were not affected by treatment. Soil test N and the FWMC and load of NO (NO + NO) were significantly greater in the AC treatment, but the FWMC and load of NH were greater in the PF treatment. Loads of total dissolved N (TDN) and total N (TN) were not affected by treatment, although the concentrations of TDN and TN were greater in the AC treatment. The PF treatment significantly increased FWMCs and loads of total dissolved P (TDP) and total P (TP). On an annual snowmelt runoff basis, the PF treatment increased the FWMC of TDP by 53% and TP by 52% and increased the load of TDP by 221% and TP by 160% compared with the AC treatment. The greater P and NH losses in the PF treatment were attributed mainly to nutrient release from forage residue due to freezing.
